Product Specific Information
Description:
The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain, along with gamma and delta subunits, is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. Crosslinking of TCR with immobilized UCHT1 induces intracellular signaling leading to T cell activation and proliferation.

Applications Tested:
- Pre-titrated and validated for flow cytometric analysis of normal human peripheral blood cells
- Recommended use: 4 µL (0.6 µg) per 100 µL test volume
- Suitable for 10⁵–10⁸ cells/test
NovaFluor Compatibility Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
Staining Recommendations:
Each NovaFluor conjugate or kit is shipped with CellBlox Blocking Buffer.
- Add 5 µL per sample when staining with NovaFluor conjugates
- For single-color controls: add 5 µL per 100 µL of cell sample (10³–10⁸ cells)

Target Information
The CD3 subunit complex transduces antigen-recognition signals into T cell cytoplasm and regulates TCR surface expression.
The complex includes CD3 gamma, delta, epsilon, and zeta subunits, all members of the immunoglobulin superfamily encoded on human chromosome 11.
Biological Roles:
- Mediates TCR-induced activation, proliferation, and survival
- Present on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes
- Absent on B and NK cells
- CD3 gene defects are associated with CD3 immunodeficiency
