Target Information

PTEN was identified as a tumor suppressor that is mutated in a large number of cancers at high frequency.
The PTEN gene encodes a phosphatidylinositol-3,4,5-trisphosphate 3-phosphatase containing a tensin-like domain and a catalytic domain similar to that of dual specificity protein tyrosine phosphatases.
Unlike most protein tyrosine phosphatases, PTEN preferentially dephosphorylates phosphoinositide substrates and negatively regulates intracellular levels of phosphatidylinositol-3,4,5-trisphosphate.
It functions as a tumor suppressor by negatively regulating the AKT/PKB signaling pathway.
PTEN modulates the AKT-mTOR signaling pathway, influencing neuron positioning, dendritic development, and synapse formation during adult neurogenesis.
Additionally, PTEN may act as a negative regulator of insulin signaling and glucose metabolism in adipose tissue.
The nuclear monoubiquitinated form possesses greater apoptotic potential, whereas the cytoplasmic nonubiquitinated form induces less tumor suppressive ability.
