Target Information
TPSB2는 단백질 코딩 유전자로, trypsin-like serine protease 계열인 peptidase family S1에 속하는 tryptase 패밀리에 포함됩니다. Tryptase는 heparin-stabilized tetramer 형태에서만 효소 활성을 가지며, 알려진 내인성 protease 억제제에 저항성을 보입니다. 여러 tryptase 유전자는 염색체 16p13.3에 클러스터되어 있으며, 3’ UTR의 높은 보존성과 5’ flank 및 3’ UTR의 tandem 반복 서열을 특징으로 합니다. 이러한 반복 서열은 mRNA 안정성 조절에 관여하는 것으로 알려져 있습니다. 또한 이 유전자는 번역 개시 Met 코돈 바로 상류에 intron을 포함하고 있으며, 이는 tryptase 유전자의 특징적인 구조입니다. TPSB2의 allele은 높은 서열 변이를 보이며, 과거에는 beta II 및 beta III 두 개의 별도 유전자로 오인된 바 있습니다. Beta tryptase는 주로 mast cell에서 발현되고, basophil에서는 alpha tryptase가 우세하게 발현됩니다. Tryptase는 천식 및 기타 알레르기·염증성 질환의 병태생리에 관련된 매개체로 알려져 있습니다.
