The FF3399 monoclonal antibody recognizes human aryl hydrocarbon receptor (AHR).
AHR is a ligand-activated transcription factor that mediates the toxic effects of environmental contaminants such as PCBs and TCDD. It also binds naturally occurring compounds in foods and metabolites. AHR resides in the cytoplasm complexed with HSP90, p23, and XAP2/AIP/ARA9. Upon ligand binding, it translocates to the nucleus, dimerizes with ARNT, and binds to the GCGTG consensus sequence in promoter regions of genes like CYP1A1.
AHR is expressed in many cell types, with highest expression in liver, and is involved in Treg and Th17 cell regulation.

Applications Tested:
- Pre-titrated and tested for intracellular staining of Th17-polarized normal human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) following BestProtocols® Protocol A.
- Recommended usage: 5 µL (0.5 µg) per test (100 µL final volume).
- Compatible with 633 nm red laser detection.
- Filtration: 0.2 µm post-manufacturing filtered.

Target Information
AHR (Ah Receptor) belongs to a protein family including ARNT (HIF-1 Beta) and Drosophila PER and SIM proteins.
It contains an N-terminal PAS domain (~200 aa) and binds DNA enhancer sequences to initiate transcription of cytochrome P-450 (CYP1A1).
AHR regulates xenobiotic-metabolizing enzymes and binds aromatic hydrocarbons such as polycyclic and halogenated compounds (e.g., TCDD).
In non-ligand activated cells, AHR complexes with HSP90 in the cytoplasm. Upon ligand binding, it transforms into a nuclear DNA-binding form, forming a heterodimer with ARNT.
Diseases associated with AHR include eosinophilic fasciitis and seborrheic dermatitis.
