Target Information
ERN1 serine/threonine kinase (endoplasmic reticulum to nucleus signaling 1) is a human homologue of the yeast Ire1 gene product. It possesses intrinsic kinase and endoribonuclease activities and plays a key role in ER-based stress signal transduction.
IRE1 acts as a sensor of unfolded proteins in the ER and initiates the unfolded protein response by splicing XBP1 mRNA, converting it into a potent transcriptional activator.
Null mutants are viable under normal conditions but essential under stress conditions that cause unfolded proteins to accumulate in the ER. Depletion of IRE1p induces apoptosis under such stress conditions.
