Product Specific Information
- For IHC (PFA fixed): Incubate antibody for 30 minutes at room temperature.
- Staining of formalin-fixed tissues requires heating sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
MMP9 (matrix metallopeptidase 9, also known as GELB or CLG4B) is a zinc- and calcium-dependent endopeptidase that degrades extracellular matrix proteins. It is secreted as a 92 kDa zymogen, with the active enzyme at 82 kDa after cleavage. MMP9 contains:
- A gelatin-binding domain (three fibronectin type II units)
- A catalytic domain with a zinc-binding site
- A proline-rich type V collagen-homologous domain
- A hemopexin-like domain
MMP9 is expressed in monocytes, macrophages, neutrophils, keratinocytes, fibroblasts, osteoclasts, and endothelial cells. It plays roles in:
- Inflammatory responses
- Tissue remodeling and wound healing
- Tumor growth and metastasis
MMP9 contributes to skin carcinogenesis and degrades type IV and V collagens. Studies indicate involvement in IL-8-induced mobilization of hematopoietic progenitor cells and tumor-associated tissue remodeling. Elevated MMP9 levels are associated with idiopathic atrial fibrillation and aortic aneurysm progression.
