Description:
The RPA-T8 monoclonal antibody reacts with the human CD8a molecule (~32–34 kDa), expressed as a heterodimer (CD8α/β) or homodimer (CD8α/α). CD8a is found on most thymocytes, a subset of mature T cells, and NK cells. It binds MHC class I and associates with p56lck kinase, playing a role in T-cell development and activation.

Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometry using normal human peripheral blood cells. Recommended ≤0.25 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize titration for best performance.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family. It plays a key role in T cell signal transduction, differentiation, and activation. CD8 is mainly expressed on T cells as a disulfide-linked heterodimer (CD8α/β), functioning as a co-receptor for MHC class I molecules.
CD8α is essential for MHC-I binding and is also found on subsets of T cells, NK cells, monocytes, and dendritic cells as homodimers (CD8α/α).
Upon MHC-I/peptide complex ligation, CD8 recruits Lck kinase, promoting lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs are crucial for pathogen and tumor cell clearance. Differentiation into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
