Target Information
Protein p21 is a tumor suppressor protein that inhibits Cyclin-dependent kinases (CDK) and is transcriptionally activated by p53. It plays a key role in the cellular response to genotoxic stress and is a major transcriptional target of p53. Nuclear p21 binds and inhibits Cdk1 and Cdk2, blocking transition from G1 to S phase or G2 to M phase after DNA damage, allowing DNA repair. In the cytoplasm, p21 has an anti-apoptotic effect by binding and inhibiting caspase 3, ASK1, and JNK. Thus, p21 exhibits dual roles in carcinogenesis, acting both as a tumor suppressor and as an oncogene depending on context.
