Target Information
Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family. It plays a critical role in signal transduction, T cell differentiation, and activation. CD8 is predominantly expressed on T cells as a disulfide-linked heterodimer of CD8α and CD8β, functioning as a co-receptor with T cell receptor (TCR) for MHC class I molecules. In contrast, CD4 acts as a co-receptor for MHC class II molecules.
CD8α is essential for binding to MHC-I and is also expressed on subsets of T cells, NK cells, monocytes, and dendritic cells as CD8α homodimers. Interaction of MHC-I/peptide complexes with CD8 triggers recruitment of Lck kinase, leading to lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs are key in pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
