Product Specific Information

This FFA21 monoclonal antibody reacts with mouse interleukin-21 (IL-21), a 17 kDa immunomodulatory cytokine mainly produced by NKT, Th17, and TFH cells.
In TFH cells, IL-21 expression triggers autocrine signaling through IL-21R and STAT3, activating Bcl6 transcription. IL-21 is essential for TFH effector function, B cell differentiation into plasma and memory cells, IgG production, and apoptosis induction in naive B cells.

In Th17 cells, IL-21 signaling through STAT3, IRF4, and RORγt upregulates IL-23R, supporting Th17 maturation and maintenance. IL-21 also downregulates Foxp3. Elevated IL-21 levels are observed in colitis models, whereas IL-21-deficient mice are resistant to chemically induced colitis.

Preliminary data suggest that clone FFA21 recognizes a different epitope than anti-mouse IL-21 clone mhalx21 (Product #51-7213-83).

Applications Reported: Suitable for intracellular staining and flow cytometric analysis.
Applications Tested: Validated using Th17-polarized mouse splenocytes restimulated with PMA, ionomycin, and brefeldin A for 5 hours, following the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00) and standard protocol available at www.thermofisher.com/flowprotocols.
Recommended usage: ≤ 1.0 µg per test (100 µL final volume; 10⁵–10⁸ cells/test). Optimize concentration for best performance.

Fluorochrome Information:
eFluor 450 is an alternative to Pacific Blue™, excited by the violet laser (405 nm) and emitting at 446 nm. Ensure instrument compatibility.


Target Information

IL-21 is a 17 kDa cytokine produced by NKT, Th17, and TFH cells. It mediates autocrine signaling via IL-21R and STAT3, promoting Bcl6 activation and TFH cell function. IL-21 regulates B cell differentiation, IgG production, and apoptosis in naive B cells.
In Th17 cells, IL-21 enhances IL-23R expression via STAT3, IRF4, and RORγt, while suppressing Foxp3. IL-21-deficient mice show resistance to colitis, whereas elevated IL-21 levels are found in inflammatory models.
