Product Specific Information

This NGZB monoclonal antibody reacts with mouse Granzyme B, a member of the granzyme serine protease family located in the granules of cytotoxic T cells and NK cells. Granzyme B plays a key role in apoptosis induction via mannose 6-phosphate receptor-mediated pathways.

  • Applications Reported: Intracellular staining followed by flow cytometric analysis
  • Applications Tested: Tested with stimulated mouse splenocytes using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00)
  • Recommended Protocol: BestProtocols® - Protocol A
  • Usage: ≤ 0.25 µg/test (100 µL final volume, 10⁵–10⁸ cells/test)
  • Excitation/Emission: 488–561 nm / 607 nm; suitable for Blue, Green, and Yellow-Green lasers
  • Filter Recommendation: 610/20 band pass filter (PE-Texas Red® equivalent)
  • Light Sensitivity: Protect from light to prevent photo-induced oxidation

Target Information

Granzyme B is a serine protease found in cytotoxic T lymphocytes (CTLs) and NK cells. It induces apoptosis through caspase activation and mitochondrial pathways. Granzyme B interacts with mannose 6-phosphate receptor as a death receptor during cytotoxic T cell-induced apoptosis. It cleaves caspase-3 and Bid protein to trigger DNA fragmentation and cell death. Various granzymes (A–G) have been identified in mouse CTLs and NK cells.
