Product Specific Information

Description:
The XMG1.2 antibody reacts with mouse interferon (IFN) gamma and is a neutralizing antibody. Mouse IFN gamma is a 20 kDa cytokine produced by activated T, B, and NK cells, exhibiting antiviral and antiparasitic activity. It acts synergistically with other cytokines such as TNF alpha and modulates immune responses through IFN gamma receptors. Glycosylation does not affect its activity.

Applications Reported:
Reported for use in intracellular staining followed by flow cytometric analysis.

Applications Tested:
Tested by intracellular staining and flow cytometry of stimulated mouse splenocytes using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).
Recommended use ≤ 0.25 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Antibody titration recommended for optimal results.

Fluorophore Information:
PE-eFluor® 610 is excited by 488–561 nm lasers and emits at 607 nm. Use a 610/20 band pass filter (equivalent to PE-Texas Red®). Protect from light due to photo-induced oxidation sensitivity.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark. Clone-specific performance should be determined empirically.

Excitation / Emission / Laser:
488–561 nm / 607 nm / Blue, Green, Yellow-Green Lasers
Filtered post-manufacturing (0.2 µm).

Target Information

IFN gamma (Interferon gamma, Type II interferon):
A macrophage activation factor primarily produced by T-lymphocytes and NK cells in response to antigens and cytokines. It is a glycoprotein homodimer (~45 kDa) with species-specific activity. Functions include antiviral and antiproliferative effects, MHC induction, macrophage activation, and enhanced B cell immunoglobulin secretion.
Human IFN gamma does not cross-react with mouse IFN gamma.
Activation occurs via IFN gamma receptor I and II, triggering the JAK-STAT pathway.
Mutations in IFN gamma gene are associated with aplastic anemia.
