Product Specific Information
The 4S.B3 monoclonal antibody reacts with human interferon-gamma (IFN-γ), a 17 kDa cytokine produced by activated T and NK cells with antiviral and antiparasitic activity. It synergizes with TNF-α to inhibit proliferation of normal and transformed cells and exhibits immunomodulatory effects on various cell types expressing IFN-γ receptors.
Applications Reported:
Intracellular staining followed by flow cytometric analysis.
Applications Tested:
Pre-titrated and validated using intracellular staining and flow cytometry of stimulated human peripheral blood cells with the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00) following BestProtocols® (Protocol A).
Recommended use: 5 µL (0.5 µg) per test in a final volume of 100 µL (10⁵–10⁸ cells/test).
Fluorophore Information:
PE-eFluor® 610 is excited by 488–561 nm lasers and emits at 607 nm. Use a 610/20 band pass filter (PE-Texas Red® equivalent). Protect from light due to photo-induced oxidation sensitivity.
Fixation:
Samples may be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal signal loss.

Target Information
Interferon gamma (IFN-γ, Type II interferon) is a macrophage activation factor produced mainly by T-lymphocytes and NK cells in response to antigens and cytokines. It functions as a homodimer (~45 kDa) with antiviral, antiproliferative, and immunomodulatory activities, including induction of MHC I/II, macrophage activation, and enhanced antibody secretion by B cells. Human IFN-γ is species-specific and does not cross-react with mouse IFN-γ.
Mutations in the IFN-γ gene are associated with aplastic anemia.
