Product Specific Information

The UCHT2 monoclonal antibody reacts with human CD5, a 67 kDa protein expressed by most thymocytes, mature T cells, and a subset of B cells. CD5 signaling activates T cells and interacts with CD72 on B cells, playing an important role in T-B cell interaction and proliferation.
This antibody is validated for flow cytometric analysis and is pre-diluted for use at 5 µL (0.25 µg) per test, defined as staining a cell sample (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorophore Information:
Super Bright 645 is a tandem dye excited at 405 nm and emits at 645 nm. Recommended detection using a 660/20 bandpass filter. Ensure the instrument can detect this fluorochrome.

Usage Notes:

• When using multiple Super Bright dyes, apply Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.
• Protect vials and stained samples from light (photo-induced oxidation).
• Samples may be fixed using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark.
• Super Bright Polymer Dyes are sold under license from Becton, Dickinson and Company.

Target Information

CD5 is a 67 kDa single-chain transmembrane glycoprotein found on all mature T lymphocytes, most thymocytes, and many T-cell leukemias and lymphomas. It also reacts with a subset of activated B cells and may regulate T-cell proliferation. CD5 is expressed on 95% of thymocytes and 72% of peripheral blood lymphocytes, with strong reactivity in T cell areas of lymph nodes.
Diseases associated with CD5 dysfunction include thymus cancer and Richter’s Syndrome.
