Product Specific Information

The RPA-T4 monoclonal antibody reacts with human CD4, a 59 kDa cell surface receptor expressed by thymocytes, T-helper cells, and monocytes. CD4 functions as a receptor for HIV, and RPA-T4 blocks HIV binding and mixed lymphocyte reactions.
RPA-T4 and OKT4 recognize different epitopes and do not cross-block each other.

Applications Tested: Pre-titrated and tested on normal human peripheral blood cells at 5 µL (0.5 µg) per test. A test is defined as staining a cell sample in a final volume of 100 µL.

Super Bright 645 is a tandem dye excited by the violet laser (405 nm) and emits at 645 nm. Use a 660/20 bandpass filter and ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity: Protect vial and stained samples from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

CD4 antigen recognizes MHC class II molecules and acts as a co-receptor for HIV. It is expressed mainly on T-helper lymphocytes, but also on monocytes, macrophages, and dendritic cells.
At the tissue level, CD4 can be detected in thymus, lymph nodes, tonsils, spleen, and specific regions of the brain and gut.
CD4 facilitates early T-cell activation by associating with the T-cell receptor complex and tyrosine kinase Lck. It may also play a role in neuronal damage in infectious and immune-mediated CNS diseases.
Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
