Product Specific Information

The OKT8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as heterodimer (CD8αβ) or homodimer (CD8αα). It is found on thymocytes, mature T cells, and NK cells. CD8 binds MHC class I and interacts with tyrosine kinase p56lck, contributing to T-cell development and activation.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and validated for flow cytometry with normal human peripheral blood cells at 5 µL (0.125 µg) per test (100 µL total volume).

Super Bright 645 is a tandem dye excited at 405 nm and emitting at 645 nm. Recommended filter: 660/20 bandpass. Use Super Bright Complete Staining Buffer (Product # SB-4401) when combining multiple Super Bright conjugates to minimize non-specific interactions.

Light Sensitivity: Protect from light; sensitive to photo-induced oxidation.
Fixation: Samples may be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark.
Laser: Violet Laser (405 nm excitation, 645 nm emission).

Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin family involved in T cell differentiation and activation. CD8 functions as a co-receptor for MHC class I molecules and is primarily expressed on T cells as CD8αβ heterodimers. It is also found as CD8αα homodimers on subsets of T cells, NK cells, monocytes, and dendritic cells. Binding of MHC-I/peptide complexes triggers Lck-mediated signaling, leading to lymphokine production and cytotoxic T lymphocyte activation. Activated CTLs play a key role in pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
