Target Information
Immunophilins are soluble cytosolic receptors that bind immunosuppressant agents such as cyclosporin A (CsA) or FK506. Proteins that bind FK506 are FK506 Binding Proteins (FKBPs), and those that bind cyclosporin A are cyclophilins (CyP). Both CyP:CsA and FKBP:FK506 complexes inhibit calcineurin, a calcium/calmodulin-dependent phosphatase involved in T-cell activation, explaining their immunosuppressive effects.
Immunophilins function as peptidyl prolyl cis-trans-isomerases (PPIases), accelerating protein folding by catalyzing rearrangement of proline-containing proteins. Within the cyclophilin family, CyPA (also known as CyP-18) is the most abundant and ubiquitous form, found in all vertebrate tissues and particularly in T-cells.
