Target Information
CD33 is a transmembrane protein of the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit protein tyrosine phosphatases SHP-1 and SHP-2 for signal transduction and receptor regulation.
CD33 is expressed on myelomonocytic lineage cells, binds sialic acid residues in N- and O-glycans, and is a therapeutic target for acute myeloid leukemia.
It is found on granulocyte and macrophage precursors in bone marrow but not on pluripotent stem cells. CD33 serves as a marker for peripheral monocytes and helps distinguish myelogenous leukemia from lymphoid or erythroid leukemias.
Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
