Target Information
CD33 is a transmembrane protein belonging to the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit SHP-1 and SHP-2 phosphatases, mediating inhibitory signaling.
CD33 is expressed on cells of myelomonocytic lineage and binds sialic acid residues on cell surfaces. It serves as a therapeutic target for acute myeloid leukemia (AML).
In bone marrow, CD33 is found on granulocyte and macrophage precursors, but not on pluripotent stem cells. It is also expressed on peripheral monocytes and is useful for distinguishing myelogenous leukemia cells from lymphoid or erythroid leukemias.
Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
