Target Information

Isocitrate dehydrogenases catalyze the oxidative decarboxylation of isocitrate to 2-oxoglutarate. These enzymes belong to two distinct subclasses, one utilizing NAD(+) and the other NADP(+) as electron acceptors. Five isocitrate dehydrogenases have been reported: three NAD(+)-dependent (mitochondrial matrix) and two NADP(+)-dependent (one mitochondrial, one cytosolic).
The protein encoded by the IDH1 gene is the NADP(+)-dependent isocitrate dehydrogenase found in the cytoplasm and peroxisomes, containing the PTS-1 peroxisomal targeting signal sequence. This enzyme plays a role in regenerating NADPH for intraperoxisomal reductions and in peroxisomal reactions consuming 2-oxoglutarate, including alpha-hydroxylation of phytanic acid.
The cytoplasmic enzyme contributes significantly to NADPH production.
