Product Specific Information

The eBio4KB128 monoclonal antibody recognizes human CD22 (Siglec-2), a member of the siglec subgroup of the Ig superfamily. CD22 is a type I transmembrane glycoprotein composed of two polypeptide chains (CD22α and CD22β, 130 and 140 kDa) produced by alternative splicing.

CD22 is highly expressed on mature B cells and B-cell lymphomas. The extracellular portion contains seven Ig-like domains that can bind sialic acid moieties on various cell types. The intracellular region includes six tyrosine residues within ITIM and activation-like motifs, phosphorylated upon B-cell receptor engagement, regulating both positive and negative B-cell receptor signaling.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometry using normal human peripheral blood cells. Recommended usage: 5 µL (0.2 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Note:
NovaFluor dyes are incompatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI. Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for NovaFluor dyes.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining. Add 5 µL per sample to antibody cocktails before combining with cells. For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures minimizing spillover and spread impacts.

Target Information

CD22 (BL-CAM) is a type 1 integral membrane glycoprotein (130–140 kDa) expressed in the cytoplasm and membrane of B-lymphocytes. It appears early in B-cell differentiation and is expressed on mature B cells but absent in plasma cells. CD22 is strongly expressed in hairy cell leukemia and may assist in phenotyping mature leukemias.

CD22 preferentially binds α2,6-linked sialic acid. Upon ligand-induced phosphorylation, it regulates B-cell receptor signaling through interactions with Src family kinases and phosphatases. CD22 is positive in diffuse large B-cell lymphoma and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.
