Target Information

The androgen receptor (AR) is a member of the steroid-hormone receptor superfamily of nuclear receptors. The receptor (~90 kDa) contains three major functional domains: the N-terminal domain, DNA-binding domain, and androgen-binding domain.
AR is a ligand-activated transcription factor that binds testosterone (T) and dihydrotestosterone (DHT). Upon ligand binding, AR dissociates from accessory proteins, translocates to the nucleus, dimerizes, and activates transcription of androgen-responsive genes.
The AR signaling pathway is essential for the development and function of male reproductive organs (prostate, epididymis) and also affects non-reproductive tissues such as muscle, hair follicles, and brain.
AR is a phosphoprotein that regulates MAP kinase activity; inhibition of MEK1/2 correlates with AR phosphorylation changes.
Abnormal AR signaling is linked to diseases including prostate cancer, Kennedy’s disease, and male infertility. Mutations in AR cause complete androgen insensitivity syndrome (CAIS).
