Target Information
Circadian rhythmicity is a fundamental biological property observed across diverse organisms, including animals, plants, and fungi. In mammals, endogenous biological clocks are regulated by a protein-mediated autoregulatory feedback loop involving members of the bHLH-PAS (PER-ARNT-SIM) transcription factor family.
CLOCK-ARNT3 heterodimers bind to E-box regulatory elements (CACGTG) to stimulate gene transcription. PER proteins, together with TIM, negatively regulate their own transcription by inhibiting the CLOCK-BMAL1 transactivation complex, thereby maintaining circadian oscillation.
