Epithelial sodium channels (ENaC) are amiloride-sensitive members of the Degenerin/epithelial sodium channel (Deg/ENaC) superfamily of ion channels.
These channels share structural similarity, possessing two intracellular termini, two transmembrane segments, and a large extracellular loop with a conserved cysteine-rich region.
There are three homologous isoforms of ENaC (alpha, beta, and gamma). ENaC in the kidney, lung, and colon is essential for sodium and fluid balance and mediates aldosterone-dependent sodium reabsorption in the distal nephron, thereby regulating blood pressure.
ENaC function is influenced by interaction with the cystic fibrosis transmembrane conductance regulator (CFTR) chloride channel.
Gain-of-function mutations in beta- or gamma-ENaC can cause Liddle’s syndrome (hypertension), while loss-of-function mutations in alpha- or beta-ENaC can lead to pseudohypoaldosteronism type 1 (PHA-1).
