The monoclonal antibody N11 reacts specifically with the 77 amino acid form of IL-8 (IL-8₇₇, IL-8(1-77), CXCL8₇₇) and does not react with the more common 72 amino acid form.
This 77 amino acid variant, often referred to as endothelial IL-8, is secreted in cultured leukocytes. The shorter 72 amino acid form results from cleavage of an N-terminal pentamer after secretion.
IL-8 is a C-X-C chemokine involved in neutrophil chemotaxis and activation. IL-8₇₇ has reduced chemotactic activity compared to IL-8₇₂ but exhibits distinct pro-apoptotic and tumor cell chemotactic functions.
IL-8₇₇ is the predominant isoform in premature neonates due to serum protease deficiency not observed in term neonates.

Applications Reported: Suitable for immunoblotting (WB).
Applications Tested: Validated by immunoblot of peripheral blood mononuclear cells stimulated with LPS for 4 hours. Recommended concentration: 1–5 µg/mL.
Antibody should be titrated for optimal assay performance.

Target Information

Interleukin 8 (IL-8, CXCL8) is a 72 amino acid pro-inflammatory chemokine that binds to IL-8RA and IL-8RB receptors. It acts as a chemoattractant and angiogenic factor, induced by inflammatory stimuli in macrophages and endothelial cells.
In endothelial cells, IL-8 is stored in Weibel-Palade bodies. It was first isolated from osteosarcoma cells and contains the ELR motif (Glu-Leu-Arg). IL-8 signals through CXCR1 and CXCR2 receptors.
Alternative names include NAP-1, GCP-1, MONAP, and protein 3-10C. IL-8 and related chemokines form a gene cluster on chromosome 4q.
IL-8 plays roles in tumor angiogenesis and growth, as well as in the pathogenesis of bronchiolitis, a common viral respiratory disease.
