Product Specific Information
The OKT3 monoclonal antibody reacts with an epitope on the epsilon-subunit within the human CD3 complex. It exhibits potent immunosuppressive properties in vivo and has been used effectively in the treatment of renal, heart, and liver allograft rejection.
CD3 subunits (gamma, delta, epsilon) are essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. Crosslinking of TCR triggers intracellular signaling leading to T-cell activation and proliferation.
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry of normal human peripheral blood cells.
Use at 5 µL (1 µg) per test in a final volume of 100 µL (10⁵–10⁸ cells/test).

Target Information
The CD3 complex transduces antigen-recognition signals into T cells and regulates TCR surface expression. It includes subunits CD3 gamma, delta, epsilon, and zeta, encoded on human chromosome 11. These subunits have cytoplasmic tails that interact with signaling molecules via tyrosine-based motifs.
CD3 antigens are found on 68–82% of normal peripheral blood lymphocytes and 65–85% of thymocytes, but not on B or NK cells. Reduced T-cell percentages may occur in autoimmune diseases. Mutations in CD3 genes are associated with CD3 immunodeficiency.
