Product Specific Information

The FLEG monoclonal antibody recognizes human CD162 (P-selectin glycoprotein ligand-1, PSGL-1), a 120 kDa homodimer expressed on monocytes, neutrophils, granulocytes, peripheral T cells, B cells, and CD34+ hematopoietic progenitor cells. CD162 mediates leukocyte interactions with activated platelets and endothelium via binding to CD62P, CD62E, and CD62L, playing a key role in adhesion and rolling. Binding to P-selectin on HPCs can inhibit hematopoiesis, and CD162 also serves as a receptor for enterovirus 71.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells at 5 µL (0.125 µg)/test. A test is defined as staining a cell sample in 100 µL final volume, with cell numbers ranging from 10⁵ to 10⁸ cells/test.

Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Use a 610/20 bandpass filter. Verify instrument compatibility before use.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 2–8°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

CD162 (PSGL-1) is a 120 kDa homodimer found on leukocytes and platelets. It binds CD62P (P-selectin), CD62L (L-selectin), and CD62E (E-selectin) to mediate leukocyte adhesion, rolling, and extravasation. It is also an important immune marker for T cell exhaustion in chronic viral infections and cancer.
