Product Specific Information

The IT2.2 monoclonal antibody reacts with human CD86 (~80 kDa), also known as B7-2, a costimulatory molecule expressed on B cells, macrophages, and dendritic cells. CD86 interacts with T cell surface molecules CD28 and CTLA-4, playing a crucial role in T-B cell communication, costimulation, and immune response regulation.

Each product includes:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry using normal human peripheral blood cells; use at 5 µL (0.2 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.

  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls: use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, enabling engineered fluorescent signatures optimized for minimal spillover and spectral spread.

Target Information

CD86 is a ligand for CTLA4 and CD28, acting as a costimulatory molecule in T-cell activation. Insufficient co-stimulation can induce immune tolerance. CD86 is a type I membrane protein of the immunoglobulin superfamily, expressed on antigen-presenting cells. It regulates T-cell activation and immune response intensity. Alternative splicing generates multiple isoforms. CD86 dysfunction has been associated with gallbladder squamous cell carcinoma and myocarditis.
