Product Specific Information
- For IHC (PFA fixed): Incubate antibody for 30 minutes at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
MMP9 (matrix metallopeptidase 9, also known as GELB, CLG4B) is a zinc and calcium-dependent endopeptidase that degrades extracellular matrix proteins.
It is secreted as a 92 kDa zymogen, and cleavage of pro-MMP9 yields an active enzyme of 82 kDa.
MMP9 contains:
- Gelatin-binding domain (three fibronectin type II units)
- Catalytic domain with zinc-binding site
- Proline-rich type V collagen-homologous domain
- Hemopexin-like domain
MMP9 is expressed by monocytes, macrophages, neutrophils, keratinocytes, fibroblasts, osteoclasts, and endothelial cells.
It plays roles in inflammatory responses, tissue remodeling, wound healing, tumor growth, and metastasis.
Additionally, MMP9 contributes to skin carcinogenesis and degrades type IV and V collagens.
Studies suggest involvement in IL-8-induced mobilization of hematopoietic progenitor cells and tumor-associated tissue remodeling.
Elevated MMP9 levels have been linked to idiopathic atrial fibrillation and aortic aneurysm progression.
