Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 up-regulates transcription of target genes such as p21waf1, leading to G1 or G2/M arrest.

Structurally, p53 includes:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerization domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 is crucial for activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation. p53 plays an essential role in preventing proliferation of damaged cells and functions as an anticancer agent. It is also linked to aging, oxidative stress, and learning/memory processes.

Mutations in TP53 cause loss of tumor suppressor function and are frequently found in various human cancers. Germline mutations can lead to inherited cancer syndromes such as Li-Fraumeni syndrome.
