Target Information
MDM2 is a ubiquitin ligase for p53 and plays a central role in regulating the stability of p53. It is located on chromosome 12q. Akt-mediated phosphorylation of MDM2 at Ser166 and Ser186 enhances its interaction with p300, leading to MDM2-mediated ubiquitination and degradation of p53. Phosphorylation also inhibits binding to p19ARF, promoting p53 degradation. MDM2 negatively regulates p53 function by inhibiting its transactivation and promoting its degradation through E3 ubiquitin ligase activity. Overexpression of MDM2 can result in inactivation of p53 and decreased tumor suppressor function. MDM2 is overexpressed in various human malignancies, including soft tissue carcinomas and breast cancer. It also interacts with proteins such as retinoblastoma 1 and ribosomal protein L5, influencing apoptosis, cell cycle, and tumorigenesis. More than 40 alternatively spliced transcript variants have been identified in both tumor and normal tissues.
