Target Information

MDM2 is a ubiquitin ligase for p53 and plays a central role in regulating the stability of p53. It is located on chromosome 12q. Akt-mediated phosphorylation of MDM2 at Ser166 and Ser186 enhances its interaction with p300, promoting p53 ubiquitination and degradation. Phosphorylation also prevents MDM2 binding to p19ARF, further enhancing p53 degradation. Overexpression of MDM2 can inactivate p53, reducing its tumor suppressor function. MDM2 also participates in apoptosis and cell cycle regulation and is overexpressed in various human cancers, including soft tissue carcinomas and breast cancer. It interacts with proteins such as retinoblastoma 1 and ribosomal protein L5. Over 40 alternatively spliced transcript variants of MDM2 have been identified in both tumor and normal tissues.
