Product Specific Information
- For immunohistochemistry (PFA-fixed): incubate antibody for 30 minutes at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
BCL-2 is a key regulator of apoptosis that functions to either inhibit or promote cell death. Members of the BCL-2 family dimerize to modulate apoptotic activity. For example, Bag1 forms a heterodimer with BCL-2 to enhance its anti-apoptotic effect, while Bax and Bak promote cytochrome c release from mitochondria to initiate apoptosis.
Constitutive expression of BCL2, such as translocation to the Ig heavy chain locus, is implicated in follicular lymphoma. In these lymphomas, neoplastic germinal centers express high levels of BCL-2 alpha protein, unlike normal germinal centers. Two transcript variants of BCL-2 differ in their C-terminal ends. Overexpression of BCL-2 has been linked to human cancers, including B-cell lymphoma and prostate cancer.
