• Immunohistochemistry (PFA fixed):
  Incubate antibody for 30 min at room temperature.
  Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Cyclin D1 (PRAD-1, bcl-1)는 세포주기 조절에 핵심적인 단백질로, CDK4 및 CDK6과 결합하여 Rb 단백질을 인산화함으로써 세포주기 진행을 조절합니다.
Cyclin D1은 다양한 인간 종양에서 과발현되는 원암유전자(proto-oncogene)로 알려져 있으며, 특히 mantle cell lymphoma(MCL)에서 높은 발현을 보입니다.
이 단백질은 G1기 동안 합성되어 성장인자 자극에 따라 CDK4/6 복합체를 형성하며, Rb의 억제 기능을 비활성화하고 CDK 억제제(p21Cip1, p27Kip1)를 조절합니다.
Cyclin D1은 성장인자 자극이 없을 경우 불안정하며, 인산화에 의해 유비퀴틴 매개 분해를 거쳐 G1/S기 정지에 관여합니다.
Cyclin D1은 세포주기 및 암 관련 이상 연구에 유용한 표적 단백질입니다.
