Product Specific Information

The monoclonal antibody eBio8D9 reacts with an epitope shared by mouse CD21 (CR2) and CD35 (CR1).
CD21 and CD35 are alternatively spliced transcripts from the Cr2 gene, producing cell-surface proteins of 145 and 190 kDa, respectively.

  • Expressed by mature B cells, not on thymocytes, peripheral T cells, erythrocytes, or platelets
  • May also be expressed on macrophages
  • CD21 is a receptor for complement component C3d and Epstein-Barr virus (EBV)
  • In association with CD19 and CD81, CD21 participates in B-cell activation
  • Cr2-deficient mice show impaired inflammatory and humoral immune responses

The anti-mouse CD21/35 monoclonal antibody clones eBio4E3 and eBio8D9 do not cross-block each other, indicating different epitope binding.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes (≤0.25 µg/test).
A test is defined as the amount of antibody that stains a cell sample in 100 µL volume. Cell number may range from 10⁵ to 10⁸ cells/test.
Antibody titration is recommended for optimal performance.


Target Information

CD21 (Complement receptor 2, CR2) binds C3 complement fragments, especially breakdown fragments covalently attached to complement-activating surfaces or antigens.
It plays roles in:

  • Uptake and retention of immunocomplexes
  • Survival of memory B cells
  • Development and maintenance of humoral responses to T-dependent antigens

CD21 also serves as a receptor for Epstein-Barr virus and is involved in targeting prions to follicular dendritic cells, facilitating neuroinvasion after peripheral exposure.
A soluble form (sCD21) is shed from lymphocyte surfaces and retains ligand-binding ability.

CD35 encodes a membrane glycoprotein found on peripheral blood cells, glomerular podocytes, and follicular dendritic cells.
It acts as a receptor for complement components C3b and C4b and regulates complement cascade activity.
Variation in CD35 forms the basis of the Knops blood group system.

A secreted plasma form has been described, though its full-length structure remains undetermined.
