Product Specific Information

The 53-6.7 monoclonal antibody reacts with the mouse CD8a molecule, a 32–34 kDa surface receptor expressed as CD8αβ or CD8αα dimers. CD8 binds to MHC class I and associates with protein tyrosine kinase p56lck, playing a role in T cell development and activation.

• Applications Tested: Tested on mouse splenocytes; recommended ≤0.05 µg/test (100 µL final volume)
• Usage: Adjust cell number empirically (10⁵–10⁸ cells/test). Titrate for optimal performance.
• Each product includes 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.
• Compatibility: NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• Blocking Buffer: Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample or per 100 µL of cell sample for single-color controls.
• Technology: NovaFluor conjugates utilize Phiton technology for engineered fluorescence with minimized spillover and spread.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family involved in T cell signal transduction, differentiation, and activation. It forms heterodimers (CD8αβ) or homodimers (CD8αα) and serves as a co-receptor for MHC class I molecules. CD8 is expressed on cytotoxic T cells, NK cells, monocytes, and dendritic cells. Upon MHC-I/peptide binding, CD8 recruits Lck kinase, leading to cytokine production and cytotoxic T lymphocyte activation. IL-2, IL-12, and TGF-β1 enhance CTL differentiation.
