Product Specific Information

The 53-6.7 monoclonal antibody reacts with the mouse CD8a molecule, a 32–34 kDa cell surface receptor expressed as CD8αβ or CD8αα dimers.
CD8 binds to MHC class I and associates with p56lck, playing a key role in T cell development and activation.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes (≤0.5 µg/test).
A test is defined as staining in a final volume of 100 µL (10⁵–10⁸ cells/test). Antibody should be titrated for optimal performance.

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
    • Add 5 µL per sample to antibody cocktails before combining with cells.
    • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures minimizing spillover and spread.

Target Information

CD8 is a type I transmembrane glycoprotein of the immunoglobulin family, functioning as a co-receptor with TCR for MHC class I molecules.
Predominantly expressed on T cells as CD8αβ heterodimers, and also as CD8αα homodimers on subsets of T cells, NK cells, monocytes, and dendritic cells.
Engagement with MHC-I/peptide complexes triggers Lck recruitment, leading to lymphokine production and cytotoxic T lymphocyte activation.
Activated CTLs are essential for pathogen and tumor cell clearance. Differentiation of naive CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
