Target Information

The second messengers cAMP and cGMP are key regulatory molecules involved in numerous signal transduction pathways. Their levels are controlled by synthesis via nucleotide cyclases and hydrolysis by cyclic nucleotide phosphodiesterases (PDEs). PDEs form a superfamily of enzymes that convert cyclic nucleotides to corresponding nucleoside monophosphates.

PDE6 is the effector enzyme in the G protein-mediated signal transduction cascade of the visual system. It consists of multiple subunits: alpha′ (cone), alpha (rod), beta (rod), the inhibitory subunit gamma, and delta of unknown function. The catalytic core comprises alpha′/alpha′ homodimers in cones and alpha/beta heterodimers in rods. The C-terminus of both catalytic and inhibitory subunits undergoes methylation, myristoylation, and prenylation, which are critical for complex assembly and membrane association.
