Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes. After antigen retrieval, incubate samples with primary antibody for 10 minutes at room temperature.
A suggested positive control is hepatocellular carcinoma.

Target Information

Arginase-1 (Arg1) is a 35 kDa enzyme that converts L-arginine to urea and L-ornithine, the final step in the urea cycle. The resulting polyamines are crucial for cell proliferation and detoxification processes.
By degrading arginine, Arginase 1 limits NO synthase substrate availability, thereby reducing nitric oxide production.
In humans and mice, Arginase 1 is expressed in the liver, neutrophils, myeloid-derived suppressor cells (MDSCs), and neural stem cells. It is typically detected in hepatocellular carcinoma and may be expressed in tumor-infiltrating myeloid cells.
Defects in Arginase 1 cause argininemia, an autosomal recessive disorder associated with hyperammonemia.
