Product Specific Information
Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes.
After antigen retrieval, incubate samples with primary antibody for 30 minutes at room temperature.
A suggested positive control is placenta tissue.
Target Information
BIM belongs to the BCL-2 protein family. BCL-2 family members form hetero- or homodimers and act as anti- or pro-apoptotic regulators involved in various cellular activities.
The protein encoded by this gene contains a Bcl-2 homology domain 3 (BH3). It interacts with other BCL-2 family members, including BCL2, BCL2L1/BCL-X(L), and MCL1, acting as an apoptotic activator.
Expression of this gene can be induced by nerve growth factor (NGF) and the forkhead transcription factor FKHR-L1, suggesting a role in neuronal and lymphocyte apoptosis.
Transgenic mouse studies indicate that this gene functions as an essential initiator of apoptosis in thymocyte-negative selection.
Several alternatively spliced transcript variants have been identified.
