Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin family that plays a key role in signal transduction, T cell differentiation, and activation. CD8 is predominantly expressed on T cells as a disulfide-linked heterodimer of CD8α and CD8β, functioning as a co-receptor with the T cell receptor (TCR) for MHC class I molecules. In contrast, CD4 serves as a co-receptor for MHC class II molecules.

CD8 can also form homodimers of CD8α on subsets of T cells, NK cells, monocytes, and dendritic cells. Upon binding of MHC-I/peptide complexes presented by antigen-presenting cells (APCs), CD8 recruits lymphocyte-specific protein tyrosine kinase (Lck), leading to cytokine production, motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs are essential for eliminating pathogens and tumor cells. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
