Target Information
The Na⁺/H⁺ antiporter (Nhe-1) is a membrane-bound enzyme critical for pH regulation in vertebrate cells. It is inhibited by the diuretic drug amiloride and activated by growth factors, mitogens, neurotransmitters, and tumor promoters.
Nhe-1 also serves as an anchor for actin filaments via interaction with ERM proteins (ezrin, radixin, moesin), maintaining cortical cytoskeleton integrity.
It may have structural roles independent of ion exchange and is implicated in hypertension. Multiple isoforms of Nhe-1 are known.
