Target Information
Smad4 is encoded by the SMAD4 gene located on chromosome 18 in humans. It is also known as Smad family member 4, Sma- and Mad-related protein 4, or Mothers against decapentaplegic (MAD) homolog 4.
Smad4 contains N-terminal MH1 and C-terminal MH2 domains involved in DNA binding and protein interactions. Upon TGF-β or BMP ligand binding, receptor Smads (R-Smads: Smad1, 2, 3, 5, 8) are phosphorylated and form heterotrimers with Smad4 (co-Smad), which translocate to the nucleus to regulate gene expression.
Smad4 functions as a tumor suppressor by regulating target genes such as Cyclin D1 (downregulation) and collagen (upregulation), influencing cell proliferation and differentiation. Dephosphorylation controls the nucleocytoplasmic dynamics of Smads.
