Target Information
Estrogen Receptors (ER) belong to the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor functions as a ligand-activated transcription factor that, upon binding to estrogen, undergoes a conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA.
When bound to EREs, ER can regulate gene transcription positively or negatively through the recruitment of coactivator or corepressor proteins. Two main forms exist — alpha and beta — encoded by ESR1 and ESR2 genes, respectively. Alternative RNA splicing generates at least four alpha isoforms (66 kDa, 53 kDa, 47 kDa, 35 kDa).
Estrogen receptors are widely expressed in various tissues, essential for sexual development and reproductive function, and also influence bone physiology. They are implicated in diseases such as breast cancer, endometrial cancer, and osteoporosis.
