Product Specific Information

The 236A/E7 antibody reacts with human Foxp3 protein (FORKHEAD BOX P3, SCURFIN, JM2). Foxp3 is a 49–55 kDa transcriptional regulator, identified as the gene defective in scurfy mice. High expression of Foxp3 is found in CD4+CD25+ regulatory T cells (Treg), and ectopic expression imparts a Treg phenotype.

Intracellular staining of human PBMCs with the 236A/E7 antibody using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523) reveals staining of the CD4+CD25bright population.

The epitope recognized by 236A/E7 differs from PCH101 (Product #72-5776). This antibody also recognizes rhesus macaque, sooty mangabey, and cynomolgus macaque.

Target Information

FOXP3 (Forkhead box protein 3) is a transcriptional regulator essential for immune homeostasis. It is 381 amino acids long and constitutively expressed in CD4+CD25+ regulatory T cells. FOXP3 defects cause the scurfy phenotype in mice and contribute to human IPEX syndrome (immune dysfunction, polyendocrinopathy, enteropathy, X-linked). FOXP3 expression is regulated by p53 in response to DNA damage in certain cancer cells.
