Product Specific Information

The monoclonal antibody 3E4 reacts with human and mouse interferon regulatory factor 4 (IRF4), a 52 kDa transcription factor involved in B cell, T cell, and macrophage function.

  • In B cells, IRF4 is highly expressed in mature plasma cells and regulates differentiation and expression of genes such as Prdm1 (Blimp1).
  • In T cells, IRF4 contributes to Treg, Th2, Th9, and Th17 cell development, inducing Blimp1 and IL-10 expression.
  • IRF4 also plays roles in macrophage polarization and is upregulated in various hematologic malignancies.

Applications Reported:

  • Intracellular staining followed by flow cytometric analysis.

Applications Tested:

  • Tested by intracellular staining and flow cytometric analysis of stimulated normal human peripheral blood cells using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).
  • Recommended usage ≤0.5 µg per test (100 µL final volume).
  • Optimal antibody concentration should be titrated per assay.

Filtration: 0.2 µm post-manufacturing filtered.


Target Information

The IRF (interferon regulatory factor) family of transcription factors contains a conserved tryptophan pentad repeat DNA-binding domain.

  • Regulates interferon response and interferon-inducible gene expression.
  • IRF4 is lymphocyte-specific and modulates TLR signaling, essential for innate and adaptive immune activation.
  • Chromosomal translocation t(6;14)(p25;q32) involving IRF4 and IgH loci is associated with multiple myeloma.
  • Alternative splicing variants have been identified.
