Product Specific Information

Description:
The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha). CD127 complexes with CD132 (common gamma chain) to form the IL-7 receptor (IL-7R). This type I glycoprotein (75–80 kDa) is expressed on immature B cells, thymocytes, and most mature T cells, with transient down-regulation upon activation. IL-7 binding triggers Jak/STAT pathway signaling essential for lymphocyte development and T-cell homeostasis.

CD127 expression is down-regulated on CD4⁺CD25⁺ regulatory T cells (Tregs). The CD4⁺CD25⁺CD127^low population is highly enriched for regulatory T cells expressing Foxp3 and suppressive activity in vitro.

Binding of eBioRDR5 to PBMCs is blocked by pre-incubation with recombinant human IL-7 (Product #14-1079-80).

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested for flow cytometry on human peripheral blood cells. Use 5 µL (0.125 µg) per test (100 µL final volume).

Fluorochrome Information:
PerCP-eFluor™ 710 emits at 710 nm and is excited with the blue laser (488 nm). Recommended filter: 710/50 bandpass (695/40 acceptable). Protect from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal effect on brightness.

Target Information

CD127 (Interleukin-7 Receptor alpha, IL7Rα) is a glycoprotein critical for lymphopoiesis. It forms a functional receptor with the IL-7R gamma chain, mediating IL-7–induced proliferation of precursor B cells, thymocytes, and T cells. Dysregulation of CD127 is associated with severe combined immunodeficiency (SCID) and related T/B/NK cell deficiencies.
