Product Specific Information
The 4S.B3 monoclonal antibody reacts with human interferon-gamma (IFN-γ), a 17 kDa cytokine produced by activated T and NK cells with antiviral and antiparasitic properties.
It is suitable for intracellular staining followed by flow cytometric analysis.
This antibody has been pre-diluted and tested using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).
Recommended usage: 5 µL (0.5 µg) per test (100 µL final volume). Cell count: 10⁵–10⁸ cells/test.
Fluorochrome characteristics:
Brilliant Violet™ 650 (BV650) emits at 649 nm and is excited by a 405 nm violet laser. Ensure cytometer compatibility.
Staining considerations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product #SB-4401-42) or Brilliant Stain Buffer™ (Product #00-4409-75) to reduce non-specific interactions.
Fixation and storage:
- Protect from light (photo-sensitive dye).
- Fixed samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C.
- BV650 is not compatible with methanol-based fixation.

Target Information
IFN gamma (Interferon gamma, Type II interferon)
- Produced by T-lymphocytes and NK cells in response to antigens and cytokines
- Functions: macrophage activation, antiviral and antiproliferative activity, MHC induction, and immunoglobulin secretion enhancement
- Structure: homodimer of two 146 amino acid subunits (~45 kDa)
- Highly species-specific; human IFN-γ does not cross-react with mouse
- Regulated by IL-2, FGF, EGF (upregulation) and vitamin D3, DMN (downregulation)
- Gene mutations associated with aplastic anemia
