Product Specific Information

MA1-064 detects assembly polypeptide 2 (AP2) from a wide variety of mammalian sources, including human, hamster, monkey, bovine, rat, and mouse.
It recognizes the products of both alpha-adaptin genes (alpha A and alpha C) as well as an alternatively spliced isoform found in neurons.

MA1-064 has been successfully used in Western blot, immunofluorescence, immunocytochemistry, and immunoprecipitation procedures.
By Western blot, this antibody detects an ~100 kDa doublet representing the two isoforms of alpha adaptin in rat brain (alpha C is the dominant form).

Microinjection of cells with MA1-064 results in variable blockade of endocytosis.
The MA1-064 immunogen is purified adaptors.

Target Information

Adaptor protein complex 2 (AP-2) is involved in clathrin-dependent endocytosis.
AP-2 functions as a cargo receptor that selectively sorts membrane proteins involved in receptor-mediated endocytosis.
It plays a role in recycling synaptic vesicle membranes and recognizes endocytosis signal motifs (Y-X-X-[FILMV] and [ED]-X-X-X-L-[LI]) within cytosolic tails of transmembrane cargo molecules.

AP-2 alpha subunit binds polyphosphoinositide-containing lipids, positioning AP-2 on the membrane and acting as a scaffolding platform for endocytic accessory proteins.
Both AP-2 alpha and sigma subunits contribute to cargo recognition and vesicle formation.
