Description:
The 122 monoclonal antibody reacts with mouse B7-DC (PD-L2), a member of the B7 family with a molecular weight of ~25 kDa. It is primarily expressed by dendritic cells and monocytes/macrophages in mice. Unlike other B7 family members, B7-DC binds PD-1 rather than CD28/CTLA-4, influencing T cell proliferation depending on activation state. Clone 122 blocks binding of TY25, another anti-mouse B7-DC antibody.

Validated for flow cytometric analysis.
Applications Tested:
Tested on mouse bone marrow-derived dendritic cells at ≤0.5 µg/test (100 µL final volume). Optimal titration is recommended.

Programmed death-ligand 2 (PD-L2, B7-DC):
A B7 ligand family member that binds PD-1, acting as an inhibitory immune checkpoint molecule. Expressed mainly in dendritic cells but inducible in various cell types by cytokines such as IL-4, TNF-α, and IFN-γ. PD-L2 has a higher affinity for PD-1 than PD-L1 and modulates T cell activation and cytokine secretion. It also binds RGMb, influencing respiratory tolerance through BMP/neogenin signaling pathways. The human PD-L2 monomer comprises a 201-aa extracellular domain, 21-aa transmembrane domain, and 32-aa cytoplasmic domain.
